Cognitive research peptides act principally through upregulation of brain-derived neurotrophic factor (BDNF), nerve growth factor (NGF), and related neurotrophins, or through direct receptor agonism at hepatocyte growth factor (HGF)/c-Met. The pathway converges on hippocampal synaptic plasticity, neurogenesis, and neuronal survival. Includes Russian-registered Semax and Selank, the multi-country-licensed multi-component cerebrolysin, and the pre-clinical HGF/c-Met potentiator Dihexa.

Cognitive and nootropic peptide research addresses the neurotrophic signalling axis — the neurotrophin/Trk-receptor pathway (BDNF/TrkB, NGF/TrkA) and adjacent growth-factor systems (HGF/c-Met) — that supports adult hippocampal neurogenesis, long-term potentiation, dendritic spine formation, and the structural correlates of learning and memory. Reduced neurotrophin signalling is a feature of major depression, Alzheimer's disease, Parkinson's disease, post-traumatic brain injury, and the broader cognitive-decline phenotype of aging. Pharmacological strategies that upregulate endogenous neurotrophin expression bypass the blood-brain-barrier delivery problem of exogenous recombinant neurotrophin protein replacement — the practical rationale for the research peptides in this category. The pharmacological space is organised around four distinct mechanistic strategies. Semax (an ACTH 4-10 analogue, Met-Glu-His-Phe-Pro-Gly-Pro, MEHFPGP) upregulates hippocampal BDNF and NGF expression through mechanisms involving melanocortin-receptor-mediated transcriptional effects; the intranasal delivery route bypasses BBB limitations via olfactory and trigeminal pathways, achieving CSF concentrations 10-50 fold higher than equivalent IV doses. Selank (a tuftsin analogue, TKPRPGP, developed by the same Russian Institute of Molecular Genetics that produced Semax) modulates enkephalinase activity and produces principally anxiolytic effects through GABAergic and serotonergic system modulation, with secondary BDNF effects. Both are registered as prescription medicines in Russia and CIS countries but unlicensed in the UK. Cerebrolysin is mechanistically distinct — a multi-component preparation of porcine brain-derived peptides and amino acids produced by controlled enzymatic hydrolysis. It contains ~25% peptides below 10 kDa (with functional mimicry of NGF, BDNF, GDNF, CNTF) and ~75% free amino acids, with additional pleiotropic effects on amyloid precursor protein processing, neuroinflammation, and synaptic protein synthesis. Registered as a prescription medicine in 50+ countries (most of continental Europe, Russia, China, India) for stroke, traumatic brain injury, vascular dementia, and Alzheimer's disease indications. The Western regulatory framework has been more cautious — the CASTA acute stroke Phase III trial (Heiss et al., Stroke 2012) did not meet its primary endpoint, though post-hoc analyses suggested moderate-severity subgroup benefit. Not licensed in the UK. Dihexa (PNB-0408, N-hexanoic-Tyr-Ile-(6)-aminohexanoic amide) engages an entirely separate mechanism — HGF/c-Met potentiation at femtomolar concentrations (~10⁻¹⁵ M), approximately seven orders of magnitude more potent than BDNF on equivalent dendritic-spine-formation readouts in hippocampal slice preparations (McCoy et al., J Pharmacol Exp Ther 2013). The mechanism is fundamentally different from the BDNF/NGF-upregulating cognitive peptides. Additionally, Dihexa achieves meaningful oral bioavailability through its N-terminal hexanoyl modification — the practical advantage over Semax and Selank which require intranasal delivery. Standard pre-clinical cognitive research endpoints include hippocampal BDNF and NGF expression by ELISA and qRT-PCR; behavioural learning and memory batteries (Morris water maze, novel object recognition, contextual fear conditioning); electrophysiological hippocampal CA3-CA1 long-term potentiation in slice preparations; dendritic spine density quantification by Golgi staining or two-photon imaging; anxiolytic paradigms (elevated plus maze, light/dark box) for the Selank pharmacology dimension; and validated clinical cognitive batteries (ADAS-Cog for Alzheimer's research, MoCA for general cognitive screening, neuropsychological testing batteries for healthy-subject nootropic research). UK regulatory status is uniform across the class — none is MHRA-licensed. Cerebrolysin has widespread European registration and is theoretically accessible through the unlicensed-medicines 'specials' route but is rarely pursued given the absence of UK clinical-prescribing infrastructure familiar with cerebrolysin. Semax and Selank have Russian/CIS prescription status not translated to the UK. Dihexa is pre-clinical only. None currently sits on the WADA Prohibited List as specifically named substances, though athletes should verify the current annual Prohibited List. Open research questions include comparative human efficacy of intranasal Semax versus oral Dihexa for cognitive endpoints, the clinical relevance of cerebrolysin's mixed CASTA Phase III result versus its extensive real-world use record, and whether HGF/c-Met potentiators can achieve human clinical validation in Alzheimer's disease indications where the related fosgonimeton has produced mixed Phase II results.
